Small nucleolar RNA SNORD81 (SNORD81)

Target
SNORD81
Molecular classification
Small nucleolar RNA (snoRNA), Non-coding RNA, Box C/D snoRNA (SNORD), RNA guide molecule
01

Overview

Small nucleolar RNA SNORD81 is a member of the box C/D class of snoRNAs containing the conserved motifs C (UGAUGA) and D (CUGA). It functions as a guide RNA for 2′-O-methylation of other RNAs, most notably rRNA, by base pairing with its targets and recruiting the methyltransferase fibrillarin within the nucleolus. SNORD81 is generally encoded in intron 11 of the GAS5 gene in mammals and participates in the maturation and modification of ribosomal RNA, which is essential for accurate ribosome assembly and function. While aberrant expression or mutation of snoRNAs broadly can contribute to disease states, specific disease associations for SNORD81 have not been clearly established. It acts as part of small nucleolar ribonucleoprotein complexes (snoRNPs) and does not possess catalytic activity itself but directs enzymatic activity through its interactions with core proteins like fibrillarin, NOP56, NOP58, and SNU13.

Mechanism of action

Not applicable (no drugs specifically target SNORD81; snoRNAs act as RNA guides rather than direct druggable entities)

03

Biological functions

Guide for 2′-O-methylation of target RNAs, especially rRNAPosttranscriptional modification of ribosomal RNAs (rRNAs)Assembly and maturation of ribosomal subunits (indirect through RNA modifications)Nucleolar localization and ribonucleoprotein complex formation with partner proteins like fibrillarin, NOP56, NOP58, SNU13
04

Disease associations

Cancer (general role for snoRNAs in tumorigenesis and metastasis; SNORD81 itself is not strongly linked to specific cancers, but broader snoRNA dysregulation is reported in various cancers)Other (possible roles in cellular transformation due to perturbations in global snoRNA function, not specifically documented for SNORD81)
05

Safety considerations

None specific to SNORD81 (snoRNAs themselves are not targeted by drugs; challenges and safety concerns are generic for RNA-based therapeutics targeting snoRNA pathways, such as off-target effects and delivery issues)
06

Interacting drugs

None known (snoRNAs, including SNORD81, are not direct targets of approved therapeutics or investigational drugs)
07

Biomarkers

General snoRNAs, including SNORD81, have been explored as potential diagnostic or prognostic biomarkers in cancer, but SNORD81 is not yet established in the clinic as a biomarkerrRNA methylation patterns regulated by C/D box snoRNAs could potentially serve as molecular biomarkers
