Small proline-rich protein 2C (SPRR2C) is a member of the small proline-rich protein family primarily expressed in epithelial tissues such as skin. SPRR2C is highly expressed in keratinocytes, especially during inflammatory responses as seen in psoriasis, and largely localizes to the cytoplasm. It functions in regulating cell proliferation, cell cycle progression, and apoptosis, as well as modulating expression of a variety of inflammatory cytokines (e.g., IL-1β, IL-6, IL-8) via the PI3K/AKT/mTOR pathway. SPRR proteins are structural components of the cornified envelope, contributing to the formation and maintenance of the skin’s protective barrier. Recent research indicates SPRR2C upregulation is associated with increased proliferation and inflammatory activity in skin disease, implicating it as a potential diagnostic and therapeutic target in conditions like psoriasis and possibly epithelial cancers[1][3][4][5].
