Overview

Small proline-rich protein 2E is a member of the small proline-rich protein (SPRR) family, clustered within the epidermal differentiation complex on chromosome 1q21[1][4]. SPRR2E is a cross-linked envelope protein expressed in keratinocytes, where it first appears in the cell cytosol and ultimately becomes cross-linked to plasma membrane proteins by transglutaminases to form the insoluble cornified envelope that underlies the cell membrane of terminally differentiated squamous epithelial cells[2][3][4]. This structure provides a critical permeability barrier for the skin, protecting against environmental insults and pathogens[1][2][4]. In addition, SPRR proteins, including SPRR2E, have been shown to have direct antimicrobial activity, contributing to cutaneous immune defense by disrupting bacterial membranes[6]. Dysregulated SPRR2E expression has been associated with skin barrier diseases such as psoriasis and ichthyosis vulgaris[1][4]. There is currently no evidence that SPRR2E is a direct therapeutic target for small molecules or biological drugs[3][4][6].
