Overview

Small proline-rich protein 2F (SPRR2F) is a structural protein encoded by the SPRR2F gene, part of the small proline-rich (SPRR) family involved in the formation of the cornified envelope in keratinocytes by acting as a cross-linking protein that contributes to the skin’s barrier function[3]. SPRR2F is minimally expressed in healthy tissues, but its expression is rapidly and strongly upregulated in response to tissue injury, especially in the kidney, skin, uterus, esophagus, and bladder[1][3]. In the kidney, SPRR2F is a robust early response gene to acute and chronic renal injury, where it plays a protective role by helping to defend against reactive oxygen species (ROS) generated during inflammation and tissue stress, thus reducing damage and supporting tissue viability[1]. In animal models, loss of SPRR2F increases susceptibility to kidney damage and oxidative stress, while its upregulation appears protective following injuries such as ischemia-reperfusion and unilateral ureteral obstruction[1]. SPRR2F is also implicated in keratinocyte differentiation and may play a broader role in epithelial responses to tissue injury[1][3]. At present, there are no known drugs targeting SPRR2F directly, nor is it classified as a core therapeutic target such as a receptor, enzyme, or transporter. Instead, it serves primarily as a structural and protective protein with potential biomarker value in renal injury[1][3].
