Overview

Small ribosomal subunit protein uS12 is an evolutionarily conserved structural component of the small (40S) eukaryotic—or 30S prokaryotic—ribosome responsible for accurate decoding of messenger RNA during translation. In humans it is encoded by RPS23; it plays a critical role in ensuring translational fidelity by participating directly at sites involved with tRNA selection and codon recognition within the mRNA channel. It also contributes to assembly processes within nucleolar pre-ribosomes ("SSU processome") that generate mature functional units required for global cellular protein synthesis. Mutations can result in developmental syndromes involving growth retardation and craniofacial abnormalities. While not currently targeted therapeutically due to its essential nature across all dividing cells, its bacterial homolog remains an important antibiotic target through agents like streptomycin that exploit differences between prokaryotic/eukaryotic forms.[1][2][4][9]

Mechanism of action

For bacterial homologs—antibiotics like streptomycin bind to uS12 on the 30S subunit causing misreading during translation initiation or elongation. Inhibitors would act by disrupting mRNA decoding or tRNA selection at the A site. No approved drugs target human uS12 directly due to essentiality for normal cell function and safety concerns.
