Smoothened (SMO) is a transmembrane protein belonging to the Class Frizzled family of G protein-coupled receptors (GPCRs). It plays a central role in the hedgehog signaling pathway, which is crucial for embryonic development and adult tissue homeostasis. SMO acts as a signal transducer regulated by Patched (PTCH). Upon activation, SMO initiates intracellular signaling events that regulate gene expression critical for developmental processes. Mutations or dysregulation of SMO are implicated in several diseases, notably cancer. Therapies like vismodegib target SMO to inhibit aberrant Hh signaling.
