Mechanism of action

SMO transduces Hedgehog signaling by relieving the inhibition of Patched (PTCH) on SMO. Drugs modulate SMO's heptahelical bundle conformation to promote or inhibit downstream signaling activity. Agonists activate SMO, while antagonists like vismodegib inhibit SMO activity.
