Overview

SNAI1 cis-regulatory enhancer RNA expressed in monocytes 1 (SCREEM1) is a type of enhancer RNA (eRNA), and more specifically a long non-coding RNA (lncRNA), transcribed from a locus near the SNAI1 gene. It forms part of the SCREEM locus, which consists of several eRNAs (including SCREEM1, SCREEM2, and SCREEM3) specifically expressed in monocytes. Activation of SCREEM1 leads to increased expression of the adjacent SNAI1 gene, a master transcriptional regulator of the epithelial-mesenchymal transition (EMT). The SCREEM-SNAI1 axis thus highlights a cis-regulatory mechanism in which non-coding RNAs mark super-enhancer regions that tightly control gene expression programs central to cell identity and behavior. While SCREEM1 itself is not established as a direct therapeutic target and its transcripts are dispensable for SNAI1 induction, its locus acts as a regulatory hub; exploration of this region may offer insights into EMT, cancer progression, immune cell biology, and potentially the pathogenesis of acute monocytic leukemia. No drugs, mechanisms of action, or biomarkers directly involving SCREEM1 have been described to date[1][2][5].
