Overview

Snake venom metalloproteinases (SVMPs) from Agkistrodon acutus (also known as Deinagkistrodon acutus) are a diverse group of zinc-dependent enzymes that play a primary role in the pathophysiology of envenomation [PMID: 15533531]. These enzymes are categorized into P-I, P-II, and P-III classes based on their domain architecture, which includes a catalytic domain and often additional disintegrin-like or cysteine-rich domains [PMID: 11163344]. SVMPs function by degrading extracellular matrix components such as type IV collagen, laminin, and fibronectin, leading to the breakdown of capillary vessel integrity and subsequent systemic hemorrhage [PMID: 16023279]. They also exhibit fibrinogenolytic activity and can interfere with platelet aggregation, contributing to severe coagulopathy in bite victims [PMID: 12628484]. While they are the primary targets for specific antivenoms, SVMPs are also studied for their potential as templates in developing cardiovascular drugs due to their potent effects on blood clotting and vessel stability [PMID: 15120210]. Therapeutic intervention typically involves the administration of polyvalent or monovalent antivenoms, which contain antibodies that neutralize the enzymatic and toxic activities of these proteins [PMID: 20546774]. Small molecule inhibitors like Batimastat have also been explored in research settings to neutralize these toxins [PMID: 11513775].
