Snf2-related CREBBP activator protein (SRCAP) is the **scaffold and ATPase subunit of the human SRCAP chromatin remodeling complex**, structurally and functionally homologous to yeast SWR1. SRCAP catalyzes **ATP-dependent histone exchange**, specifically replacing canonical histone H2A-H2B dimers with the H2A.Z-H2B variant complex, thus regulating chromatin structure and gene expression[1][3][4]. The complex does not support net nucleosome sliding but instead facilitates deposition of H2A.Z, a modification associated with transcriptional regulation and promoter activity[1][3]. SRCAP is implicated in **epigenetic control, cellular differentiation, and development**, with mutations causing Floating-Harbor syndrome (FLHS), a neurodevelopmental disorder, and involvement reported in several cancers[4]. There are currently no drugs known to directly target SRCAP, but it is being studied as a potential epigenetic enzyme and therapeutic target.
