SNW domain-containing protein 1 (SNW1) is a multifunctional nuclear protein encoded by the SNW1 gene that acts as both a transcriptional coactivator and a pre-mRNA splicing factor[1][3][6]. As a coactivator, it enhances transcription from RNA polymerase II promoters by binding the ligand-binding domains of nuclear receptors such as the vitamin D receptor, retinoid receptors, estrogen receptor, and glucocorticoid receptor, thereby promoting gene expression induced by vitamin D, retinoic acid, estrogen, and glucocorticoids[1][3]. SNW1 is also an essential component of the spliceosome, involved in both major and minor splicing complexes, and has a role in the splicing of U12-type introns and in cotranscriptional splicing[3]. It is a regulator of the NF-κB transcription factor in the nucleus, necessary for the transcriptional elongation of NF-κB target genes involved in inflammation and immune response[2]. SNW1 can modulate other pathways, including TGF-β-mediated transcription, and interacts with proteins such as SMADs and SKI oncoproteins, participating in cellular transformation and oncogenic processes[1][3]. Diseases associated with SNW1 include certain cancer syndromes and neurofibromatoses. No approved drugs are known to directly target SNW1, and there are no established roles as a biomarker or specific safety concerns reported in the literature to date.
