SOD1-DT is a divergent nuclear long non-coding RNA located at the SOD1 gene locus. It is transcribed antisense from the SOD1 promoter and can generate several transcript variants due to inclusion or exclusion of transposable elements. It is expressed in neuronal cells and can regulate the expression of SOD1 and several surrounding genes (TIAM1, HUNK, CCT8, FBXW11P1, SCAF4, CFAP298, MIS18A, URB1), likely by modulating chromatin structure or direct transcriptional regulation. SOD1-DT is upregulated during neuronal differentiation and by overexpression of the transcription factor YY1, indicating potential roles in cell fate tuning. Knockdown studies show that SOD1-DT represses the expression of multiple local genes, supporting its regulatory function. Although primarily investigated in connection with ALS and HBV infection, its exact biological function in disease remains to be fully elucidated.
