NHE1 is a ubiquitously expressed sodium/hydrogen antiporter that plays a critical role in regulating intracellular pH and cell volume. It catalyzes the electroneutral exchange of one intracellular proton (H+) for one extracellular sodium ion (Na+), contributing to cellular homeostasis, especially under conditions of acid load or cell shrinkage. Beyond its ion transport function, NHE1 acts as a scaffold for signaling complexes and interacts with cytoskeletal elements, influencing processes such as cell migration, survival, and response to mechanical stimuli. Dysregulation of NHE1 is implicated in various pathologies including cardiac hypertrophy/failure and cancer progression.
