NKCC1 is a dimeric membrane protein belonging to the SLC12 family of cation–chloride cotransporters, crucial for the electroneutral transport of Na^+, K^+, and Cl^− ions across the plasma membrane in a 1:1:2 stoichiometry. It plays essential roles in maintaining intracellular chloride concentration, regulating cell volume, and controlling neuronal excitability and epithelial secretion. Structurally, NKCC1 contains twelve transmembrane segments and operates through subtle conformational changes coordinated around conserved glycine hinge regions. It is therapeutically targeted by loop diuretics (like bumetanide and furosemide) and is implicated in a range of diseases, including epilepsy and cancer, making it a focus of ongoing drug development. NKCC1 function is regulated via phosphorylation and alternative splicing, with expression and membrane localization finely tuned in different tissue types.
