Overview

Sodium channel and clathrin linker 1 (SCLT1) is an adaptor protein that links clathrin to certain sodium channels such as Na(v)1.8 (encoded by SCN10A). It is a key component of the distal appendages of centrioles, necessary for the initiation of cilium assembly (ciliogenesis) and for regulating sodium channel function, possibly by promoting channel internalization. Mutations in SCLT1 result in impaired DAP (distal appendage) recruitment and ciliogenesis, leading to developmental defects such as craniofacial and limb abnormalities and certain ciliopathies. SCLT1 is not known to be a direct therapeutic target (e.g., no drugs target it or use it as a biomarker).
