Overview

Sodium channel proteins in parasites are integral membrane proteins that facilitate the selective passage of sodium ions (Na⁺) across cellular membranes. These channels play essential roles in maintaining ion homeostasis, generating membrane potential, and supporting various physiological processes critical for parasite survival and pathogenicity. They contribute to the acquisition of nutrients and essential ions, regulation of cell volume, generation and maintenance of membrane potential, intracellular signaling events, waste elimination, and drug extrusion mechanisms that can mediate resistance. Ion channels, including sodium channel proteins, are considered promising but underexploited drug targets against parasitic diseases due to their essential roles and unique structural features compared with host homologs. Even subtle differences (e.g., single amino acid changes) can provide selectivity for targeting parasite over host proteins.
