The thiazide-sensitive sodium-chloride cotransporter (NCC), encoded by the SLC12A3 gene, is a membrane protein located in the apical membrane of distal convoluted tubule (DCT) cells. It mediates electroneutral co-transport of sodium (Na+) and chloride (Cl-) ions from the tubular fluid into the epithelial cells. NCC plays a critical role in regulating salt balance, blood pressure, and overall electrolyte homeostasis. It is the target of thiazide diuretics, widely used antihypertensive agents. Mutations in SLC12A3 cause Gitelman syndrome, a hereditary salt-wasting disorder.
