Sodium-dependent phosphate transporter type IIb (NaPi-IIb, Npt2b, SLC34A2) is the principal transporter mediating active absorption of dietary phosphate in the small intestine. It operates via a sodium-dependent mechanism and is regulated by dietary phosphate levels and hormones such as vitamin D and FGF23. NaPi-IIb and related transporters are central to systemic phosphate homeostasis. Modulation of intestinal phosphate absorption is a key therapeutic strategy in chronic kidney disease to reduce the risk of hyperphosphatemia. Despite this, attempts to inhibit NaPi-IIb directly have not yet led to successful treatments, likely due to complex regulation and compensatory mechanisms
