Overview

Sodium-driven chloride bicarbonate exchanger (SLC4A10, also known as NBCn2 or NCBE) is a membrane protein primarily expressed in the brain, including principal and inhibitory neurons as well as choroid plexus epithelial cells[2][3][4][8]. It functions as a sodium/bicarbonate cotransporter and plays a key role in regulating intracellular and extracellular pH, neuronal excitability, and cerebrospinal fluid secretion[3][6][8]. SLC4A10 mediates the electroneutral transport of sodium and bicarbonate ions, originally thought to act as a chloride-bicarbonate exchanger, but its physiological chloride exchange is limited under normal conditions[2][3]. Disruption of SLC4A10 is associated with reduced neuronal excitability, decreased risk of seizures, and neurological deficits including epilepsy and cognitive impairment[2][3]. SLC4A10 is considered a potential therapeutic target for epilepsy and disorders involving brain pH homeostasis[2][3].
