Overview

Sodium-glucose cotransporter 2 (SGLT2), encoded by *SLC5A2*, is a high-capacity but low-affinity symporter responsible for most renal reabsorption of filtered plasma glucose. It plays an essential role in energy conservation but also contributes pathologically to hyperglycemia and kidney damage when overactive. Pharmacologic inhibition with selective agents has revolutionized management strategies for type 2 diabetes mellitus due to their combined metabolic, cardiovascular, and renoprotective benefits.
