The sodium-glucose cotransporter protein subunit 2 (SGLT2), encoded by the SLC5A2 gene, is a membrane protein primarily responsible for the reabsorption of glucose in the kidneys. Located on the apical membrane of epithelial cells lining the early segment (S1) of the proximal convoluted tubule, SGLT2 actively transports D-glucose with a sodium-to-glucose coupling ratio of 1:1. It reabsorbs approximately 80–90% of filtered glucose from glomerular filtrate back into circulation. SGLT2 is a therapeutic target for SGLT2 inhibitors (gliflozins), which are used to treat type 2 diabetes mellitus and provide cardiovascular and renal benefits.
Inhibition of renal tubular glucose reabsorption, leading to increased urinary glucose excretion and reduced blood sugar levels.
