The **sodium-hydrogen exchanger 1 (NHE1)** is a ubiquitous integral membrane protein encoded by the SLC9A1 gene in humans. NHE1 functions as an electroneutral antiporter, catalyzing the exchange of intracellular protons (H⁺) for extracellular sodium ions (Na⁺). This transporter plays a fundamental role in the regulation of intracellular pH and cell volume in virtually all mammalian cells. Structurally, NHE1 consists of a transmembrane domain responsible for ion transport and a cytosolic C-terminal domain that mediates regulation by protein interactions and phosphorylation. NHE1 also acts as a signaling hub that links membrane transport to cytoskeletal organization, cell migration, and survival, and is implicated in the pathophysiology of cardiovascular, neurological, and renal diseases. The protein is inhibited nonspecifically by amiloride and specifically by drugs such as cariporide and rimeporide. Dysregulation or inhibition of NHE1 presents therapeutic opportunities and challenges, particularly in diseases involving disrupted pH, volume regulation, or excessive cell proliferation[1][2][3][4][5][6][7].
