The sodium voltage-gated channel alpha subunit 9 (NaV1.7) is a critical component of voltage-gated sodium channels, essential for generating and conducting action potentials in excitable cells, particularly neurons. Predominantly expressed in nociceptors, it amplifies small depolarizations, enabling the neuron to reach the threshold for firing an action potential. Mutations in SCN9A can lead to altered pain perception, including congenital insensitivity to pain (loss-of-function) and pain syndromes like primary erythromelalgia (gain-of-function).
