Overview

The soft tissue collagen matrix is a fundamental structural component of the extracellular matrix (ECM), primarily composed of fibrillar proteins such as Type I and Type III collagen, which provide the essential mechanical framework and tensile strength for connective tissues (StatPearls, 2023). It serves as a dynamic environment that regulates critical biological processes, including cell adhesion, migration, and tissue repair, through biochemical and biomechanical signaling (NCBI, 2021). In various fibroproliferative disorders, such as Dupuytren's contracture and Peyronie's disease, the matrix undergoes pathological remodeling, resulting in the formation of dense, inelastic collagen cords or plaques that restrict movement and cause deformity (PubMed, 2019). This matrix is a direct therapeutic target for collagenolytic enzymes like collagenase clostridium histolyticum, which selectively hydrolyze the peptide bonds within the collagen triple helix (FDA, 2010). By breaking down these excessive collagen deposits, pharmacological intervention can restore physical function and alleviate symptoms associated with localized fibrosis.
