Overview

Soluble allergen-specific immunoglobulin G (IgG) antibodies, particularly the IgG4 subclass, are essential components of the immune system's regulatory response to environmental antigens (PMID: 21839348). They function primarily as "blocking antibodies" that compete with allergen-specific IgE for binding to allergens, thereby preventing the cross-linking of IgE on mast cells and basophils (PMID: 19538381). This competitive inhibition effectively blocks the release of inflammatory mediators like histamine and leukotrienes, which drive allergic symptoms (PMID: 26899138). Additionally, these IgG antibodies can bind to the inhibitory receptor FcγRIIb, triggering signaling pathways that suppress IgE-mediated cell activation and B cell differentiation (PMID: 24388011). In the context of therapeutic intervention, the induction of high-affinity allergen-specific IgG is the primary goal of allergen immunotherapy (AIT) for conditions such as allergic rhinitis, asthma, and venom hypersensitivity (PMID: 28212938). Monitoring serum levels of these antibodies serves as a critical biomarker for assessing the clinical efficacy and immunological progress of desensitization protocols.
