Solute carrier family 10 member 7 (SLC10A7) encodes a membrane protein localized primarily in the endoplasmic reticulum, Golgi apparatus, and plasma membrane. Despite its classification within the sodium/bile acid cotransporter family, SLC10A7 does not transport bile acids or steroid conjugates—unlike other SLC10 family members. Instead, it plays a critical role in extracellular matrix production by facilitating glycosaminoglycan and glycoprotein synthesis and trafficking, essential for bone and teeth development and proper mineralization. SLC10A7 also contributes to the regulation of intracellular calcium homeostasis. Loss-of-function mutations are associated with skeletal anomalies and glycosylation disorders, but its physiological substrates remain unknown and it lacks established pharmacological modulators.
