Overview

Solute carrier family 12 member 7 (SLC12A7), commonly known as KCC4, is a protein that mediates electroneutral potassium-chloride cotransport across the plasma membrane in a 1:1 stoichiometry, important for maintaining potassium and chloride ion homeostasis within various tissues including kidney, inner ear, nervous system, and heart[1][2][3]. KCC4 is activated by cell swelling and is crucial for processes such as basolateral chloride extrusion in the kidney, acid-base metabolism, survival of cochlear hair cells, and maintaining organ function in the inner ear[1][3][4]. Mutations affecting KCC4 function are implicated in renal tubular acidosis and can contribute to electrolyte disturbances. While related cation-chloride cotransporters are clinically targeted by diuretic drugs, there are currently no approved drugs known to specifically modulate KCC4/SLC12A7[1][4].
