Overview

GLUT1 is a facilitative uniporter responsible for the energy-independent transport of glucose across cell membranes. It is highly expressed in erythrocytes and endothelial cells lining capillaries at the blood-brain barrier, ensuring adequate glucose supply. Mutations or dysfunctions in SLC2A1/GLUT1 can cause GLUT1 Deficiency Syndrome and other neurological disorders. SLC2A1/GLUT1 overexpression supports increased glycolysis in many cancers, representing an important target for cancer research/drug development.

Mechanism of action

Passive facilitated diffusion—glucose moves down its concentration gradient without ATP consumption. The transporter alternates between outward-open and inward-open conformations allowing substrate binding/release on either side of the membrane depending on relative concentrations.
