Solute carrier family 2 facilitated glucose transporter member 6 (SLC2A6, also known as GLUT6) is a membrane protein belonging to the major facilitator superfamily (MFS) of hexose transporters. It contains 12 transmembrane domains and catalyzes the uptake of glucose and possibly other hexoses into cells[1][3]. GLUT6 is predominantly expressed in the spleen, lymphocytes, brain, and upregulated in certain cancers, contributing to enhanced glucose uptake, glycolytic metabolism, and tumor cell survival. Its intracellular targeting signals suggest a partially intracellular localization, possibly at the lysosomal membrane[3]. Overexpression of GLUT6 correlates with poor prognosis in cancers like endometrial carcinoma and glioblastoma, supporting its role in tumor aggressiveness and metabolic reprogramming[1][3]. As of now, there are no clinically approved GLUT6-specific inhibitors, but the transporter has become a subject of interest for cancer metabolic therapies.
