Overview

Solute carrier family 25 member 33 (SLC25A33) is a mitochondrial inner membrane transporter responsible for importing and exporting pyrimidine nucleotides (including uridine, thymidine, guanosine, cytosine, and inosine (deoxy)nucleoside di- and triphosphates) by an antiport mechanism[1][3]. Its primary physiological role is to supply the mitochondrial DNA and RNA synthesis machinery with necessary nucleotide precursors and to participate in mitochondrial genome maintenance and control of respiration and membrane potential[1][3]. SLC25A33 is also linked to regulation of cell proliferation and inflammatory responses, particularly in macrophage activation and sepsis, by modulating mitochondrial genome stability, ROS levels, and the cGAS-STING pathway[2][3][4][7]. Dysfunction is associated with rare human disorders (such as combined oxidative phosphorylation deficiency 8), and its modulation is being explored for targeting inflammation-related pathologies[2][3].
