Overview

Solute carrier family 35 member G1 (SLC35G1) is a transporter protein belonging to the drug/metabolite transporter superfamily and the solute carrier (SLC) family, encoded by the SLC35G1 gene[2][3][5]. It is an integral membrane protein localized predominantly at the basolateral membrane of intestinal epithelial cells, where it mediates the export of citrate into the bloodstream following its intestinal uptake[4][6]. SLC35G1 is highly expressed in the upper small intestine (duodenum and jejunum), testis, and pancreas[4][6]. Notably, its transporter activity is highly sensitive to extracellular chloride, which inhibits citrate transport—making it unique among SLC family members[4][6]. Additional proposed functions include regulation of intracellular calcium levels and homeostasis, as well as participation in nuclear export of ubiquitinated proteins via the POST system (as partner-of-STIM1)[2][3][5]. SLC35G1 has been genetically associated with various diseases, such as prostate cancer and neurodevelopmental disorders like autism spectrum disorder, but no drugs are known to directly target this transporter[2][3].
