Overview

Solute carrier family 37 member 1 (SLC37A1) is an endoplasmic reticulum membrane transporter belonging to the SLC37 sugar-phosphate/phosphate antiporter family. SLC37A1 catalyzes the exchange of glucose-6-phosphate (G6P) and inorganic phosphate (Pi) across the ER membrane, functioning as both a G6P:Pi and Pi:Pi antiporter[1][2][3][4]. It is highly homologous to certain bacterial glycerol-3-phosphate permeases and is posited to also transport glycerol-3-phosphate[3][4]. SLC37A1 is ubiquitously expressed, with transcript enrichment in kidney, bone marrow, intestine, spleen, and liver, as well as in fetal tissues and notably in neutrophils. Unlike SLC37A4 (the key G6P transporter for glucose homeostasis), SLC37A1 is not functionally coupled to glucose-6-phosphatase and does not play a known role in systemic glucose regulation. Its physiological and pathophysiological significance are not fully resolved, though increased expression in some cancers and immune cell contexts suggest emerging functional associations[1][3][4]. There are currently no recognized disease links, biomarkers, or targeted drugs for SLC37A1.

Mechanism of action

Not established/validated for known drugs. Its antiport activity (G6P:Pi exchange) would be a theoretical mechanism for antagonists, but no widely-accepted inhibitors or drug mechanisms have been described[1][2][3][4].
