Overview

Solute carrier family 4 member 1 (SLC4A1), commonly known as Band 3 or Anion Exchanger 1 (AE1), is a critical multi-pass membrane protein found predominantly in erythrocytes and the alpha-intercalated cells of the kidney (UniProt P02730). In red blood cells, it facilitates the electroneutral exchange of chloride for bicarbonate, a process essential for the efficient transport of carbon dioxide from peripheral tissues to the lungs (PubMed: 25713290). In the renal system, SLC4A1 is vital for maintaining systemic acid-base balance by mediating bicarbonate reabsorption into the blood (PubMed: 15635077). Mutations in the SLC4A1 gene are associated with several clinical conditions, including hereditary spherocytosis, Southeast Asian ovalocytosis, and distal renal tubular acidosis (OMIM: 109270). While not a primary target for many current blockbuster drugs, its function is inhibited by stilbene derivatives like DIDS in experimental settings, and it remains a key focus for research into red cell membrane stability and metabolic acid-base disturbances (PubMed: 11893319). Understanding its structure and function is essential for managing genetic anemias and renal transport disorders.
