Solute carrier family 41 member 2 (SLC41A2) is a member of the SLC41 family of magnesium transporters, homologous to the prokaryotic MgtE magnesium transporter. SLC41A2 is primarily localized to the plasma membrane and mediates trans-plasma-membrane magnesium (Mg²⁺) uptake in vertebrate cells, contributing to cellular magnesium homeostasis. Its overexpression can enhance magnesium uptake and restore proliferative capacity to cells lacking other magnesium transport mechanisms, such as TRPM7, particularly under magnesium-limited conditions. Although previously postulated to have ion channel activity based on expression in certain systems, electrophysiological studies in vertebrate cells do not confirm significant ion currents, suggesting a primarily transporter, possibly charge-neutral, mechanism. No specific disease associations, drug interactions, or clinical targeting have been established for SLC41A2, but it is essential for basic magnesium uptake and may be relevant in magnesium deficiency or perturbations of cell proliferation.
