The SLC45A2 peptide-HLA class I complex is a key immunological target in melanoma immunotherapy. SLC45A2 is a melanocyte differentiation antigen, highly and selectively expressed in many melanomas but only at low levels in normal melanocytes. Peptides derived from SLC45A2 are presented on the tumor cell surface by HLA-A*0201 or HLA-A*2402 (MHC-I) molecules and are recognized by cytotoxic T lymphocytes. Immunotherapeutic approaches—such as therapeutic cancer vaccines, adoptive T cell transfer, or TCR-engineered therapies—aim to exploit this target for selective tumor cell killing, offering improved tumor selectivity and reduced potential for harmful autoimmune reactions compared with other melanoma antigens like MART-1 and PMEL. The risk of off-tumor toxicity appears lower with this antigen, but patient monitoring for autoimmunity remains important. SLC45A2-HLA complexes are considered highly promising targets for novel T cell-based cancer immunotherapies.
Recognition of SLC45A2 peptide presented by HLA-A*0201 or HLA-A*2402 enables cytotoxic T cells to kill tumor cells expressing the corresponding complex. Adoptive transfer of SLC45A2-specific CTLs targets melanoma cells and spares normal melanocytes due to low SLC45A2 expression in healthy tissues.
