SLC7A11, also known as xCT, is a protein that functions as the light chain subunit of system Xc-, a sodium-independent cystine/glutamate antiporter. It imports extracellular cystine into cells in exchange for exporting intracellular glutamate. Cystine is then reduced to cysteine, a precursor for glutathione (GSH) synthesis, which protects cells from oxidative stress and ferroptosis. SLC7A11 is highly expressed in cancer cells, where it promotes tumor growth by enhancing antioxidant capacity. It is also involved in neurological disorders due to its role in glutamate release. SLC7A11 is regulated by transcriptional factors like NRF2 and p53, as well as epigenetic and post-translational mechanisms.
