Sonic hedgehog mRNA (SHH mRNA) (SHH mRNA)

Sonic hedgehog (SHH) mRNA is the messenger RNA transcript that encodes the Sonic hedgehog protein, a critical signaling molecule in the Hedgehog (Hh) pathway (NCBI Gene ID: 6469). This pathway plays a fundamental role in embryonic development, governing processes such as limb patterning, neural tube differentiation, and organogenesis (UniProt P48747). In adult tissues, SHH signaling is involved in stem cell maintenance and tissue repair, but its aberrant activation is strongly linked to the development and progression of various cancers, including basal cell carcinoma and medulloblastoma (Rimkus et al., 2016). Targeting SHH at the mRNA level using RNA interference (siRNA) or antisense oligonucleotides (ASOs) represents a therapeutic strategy to inhibit the pathway by preventing the synthesis of the SHH ligand itself (Katoh & Katoh, 2005). This approach is particularly valuable for treating tumors that rely on autocrine or paracrine SHH signaling and may provide an alternative for patients who develop resistance to downstream inhibitors like Smoothened (SMO) antagonists (Scales & de Sauvage, 2009).
