Overview

Sorbin and SH3 domain-containing protein 1 (SORBS1) is a multidomain adaptor protein prominent in insulin signaling, cytoskeletal organization, and cell–matrix interaction. It is an 81–142 kDa protein encoded by the SORBS1 gene on chromosome 10q23.3–q24.1, generating several isoforms by alternative splicing. SORBS1 binds to c-Cbl and vinculin, facilitating tyrosine phosphorylation and actin stress fiber formation during insulin stimulation. It regulates glucose uptake in adipose and muscle tissue, participates in focal adhesion dynamics, and acts as a tumor suppressor by limiting cell migration and invasion. Mutations or altered expression are implicated in type 2 diabetes, obesity, and the progression and aggressiveness of several cancers. SORBS1 thus represents a nexus for signal transduction, metabolic regulation, cytoskeletal architecture, and cancer biology.
