Overview

SORL1 antisense RNA 1 (SORL1-AS1), also known as 51A, is a long non-coding RNA transcribed in the antisense direction from the first intron of the SORL1 (Sortilin-related receptor 1) gene[2][3][4]. SORL1-AS1 regulates SORL1 expression and splicing, increasing levels of a non-canonical SORL1 isoform associated with reduced trafficking and altered processing of amyloid precursor protein (APP), thereby promoting amyloid-β (Aβ) accumulation. Increased expression of SORL1-AS1 is implicated in the pathogenesis of Alzheimer’s disease, making it a candidate therapeutic target and a potential biomarker for disease risk or progression[1][2][3][4]. There are currently no drugs targeting SORL1-AS1 directly, but strategies aimed at lncRNA modulation are being explored for neurodegenerative disease therapy.
