Overview

Sortilin, encoded by the SORT1 gene, is a type I transmembrane glycoprotein and a member of the vacuolar protein sorting 10 (Vps10p) family of sorting receptors. It plays critical roles in intracellular protein trafficking and cell signaling, with broad implications for metabolism, neural function, and disease. Functions as a sorting receptor within the Golgi compartment and a clearance receptor on the cell surface. It mediates transport of proteins from the Golgi to lysosomes/endosomes, and from the endoplasmic reticulum to the Golgi apparatus. It binds numerous ligands including neurotensin, BDNF precursors (proBDNF/proNGFB), apolipoprotein B100, lipoprotein lipase (LPL), among others. Variants at the SORT1 locus are associated with increased risk for coronary artery disease due to its role in lipid metabolism regulation.
