Sorting and assembly machinery component 50 homolog (SAMM50) is a conserved integral membrane protein localized in the mitochondrial outer membrane, where it forms the essential core of the sorting and assembly machinery (SAM) complex. SAMM50 is required for the insertion and folding of β-barrel proteins into the mitochondrial outer membrane, a process vital for mitochondrial biogenesis and function. SAMM50—through its 16-stranded β-barrel structure—interacts with accessory subunits Sam35 and Sam37. The complex also forms a bridge to proteins within the mitochondrial inner membrane, helping maintain cristae architecture and facilitating the correct assembly of respiratory chain complexes. SAMM50 supports diverse mitochondrial processes including protein import, membrane morphology, and metabolic homeostasis. Pathogenic variants have been linked to severe, multisystemic mitochondrial diseases, but there are no known therapeutic drugs directly targeting SAMM50 to date[1][2][3][5][6][7][9].
