Overview

Sorting nexin-10 (SNX10) is a member of the phox homology (PX) domain-containing family of phosphoinositide-binding proteins localized to endosomes, where it regulates intracellular trafficking, endosome organization, and protein localization to centrosomes and cilia. SNX10 is highly expressed in bone and gut, with important roles in bone mineral and calcium homeostasis through the regulation of osteoclastic bone resorption and gastric acid secretion. Pathogenic mutations in SNX10 are linked to autosomal recessive osteopetrosis (ARO), leading to increased bone density, bone marrow failure, and anemia. Beyond the skeleton, SNX10 has emerging roles in cancer, metabolic diseases, inflammation, and autophagy. The structure of SNX10 reveals it contains an extended PX domain distinctive from related family members, and its dysfunction has broad implications for human disease and cellular trafficking[1][2][3][4][5].
