Sorting nexin-11 (SNX11) is a member of the sorting nexin family of cytoplasmic and membrane-associated proteins characterized by the presence of a phox (PX) domain, a phosphoinositide-binding motif critical for endosomal membrane association and protein-protein interactions[1][2][3][5][6]. Unlike other family members, SNX11 does not contain additional conserved domains (such as BAR, SH3, etc.), but features an "extended PX" domain (PXe) as revealed by structural studies, including two additional C-terminal α-helices essential for its function[1]. SNX11 participates in intracellular endosomal trafficking, notably by regulating the levels of certain membrane proteins (for example, promoting the trafficking of the TRPV3 channel from the plasma membrane to lysosomes for degradation)[5][6]. SNX11 is considered a protein of unknown or incompletely defined function, and—while linked to some human diseases such as certain cancers and nerve palsies—it is not currently established as a direct therapeutic target or receptor[3][5][6].
