Sorting nexin-15 (SNX15) is a member of the sorting nexin protein family, characterized by the presence of a phox (PX) domain that binds phosphoinositides and modulates intracellular trafficking of proteins. SNX15 is involved in mediating endosomal sorting, influencing the movement of proteins from the plasma membrane to recycling endosomes or the trans-Golgi network. Overexpression of SNX15 leads to impaired maturation of certain receptors, such as the insulin and hepatocyte growth factor receptors, primarily by mislocalizing the endoprotease furin responsible for their cleavage. There are multiple transcript variants due to alternative splicing. Mutation or altered regulation of SNX15 has been associated with hereditary spastic paraplegia and multiple endocrine neoplasia, though it is not currently considered a direct therapeutic target or druggable receptor[1][2][5][7].
