Overview

Sorting nexin-19 (SNX19) is a member of the sorting nexin family, proteins involved in phosphoinositide-mediated vesicle trafficking. SNX19 localizes predominantly to the endoplasmic reticulum and functions to tether endolysosomes to the ER, thereby restricting their motility and maintaining their perinuclear positioning in cells. It contains two N-terminal transmembrane domains that anchor it to the ER, and a PX domain responsible for binding phosphatidylinositol 3-phosphate on endolysosomal membranes. Additional domains regulate its interaction with endolysosomes. In pancreatic beta cells, SNX19 helps maintain insulin-containing vesicle integrity and influences insulin secretion. Mutations or dysfunction in SNX19 are associated with disorders such as spinocerebellar ataxia and immunodeficiency, and SNX family proteins are broadly implicated in cardiovascular diseases via endocytic and signaling pathway regulation[1][2][3][4][5].
