Overview

Sorting nexin-2 is a member of the sorting nexin family, characterized by a phox (PX) domain that binds phosphoinositides, and sometimes also a BAR (Bin/Amphiphysin/Rvs) domain[2][3][4]. SNX2 is involved in intracellular protein sorting, especially within early endosomes, where it helps regulate endosomal sorting and possibly trafficking of internalized cell surface receptors such as the epidermal growth factor receptor (EGFR) to lysosomes for degradation[2][3]. Although SNX2 can bind to certain phosphoinositide species and interacts with other sorting nexins, in mammalian cells it appears to play a non-essential or minor, possibly modulatory, role in endosome-to-Golgi protein retrieval[3]. Loss of SNX2 affects protein sorting and organelle homeostasis, potentially contributing to oncogenic transformation, but current evidence does not identify it as a principal drug target or direct therapeutic receptor[1][2][3][4].
