Sorting nexin-24 is a cytoplasmic and membrane-associated protein defined by a conserved Phox homology (PX) domain that binds phosphoinositides. SNX24 is necessary for the biogenesis and maturation of platelet α-granules, localizing to endosomal compartments and α-granules during megakaryocyte development. Knockout studies demonstrate that absence of SNX24 disrupts α-granule cargo trafficking, affecting key chemokines and glycoproteins. Variants in SNX24 are associated with altered platelet traits and cardiovascular phenotypes, and, while not a therapeutic target, it participates in essential cellular trafficking mechanisms. Dysfunction of sorting nexins in general is linked to several major disease areas including neurodegenerative disease, cancer, and cardiovascular disorders.
