Sorting nexin 25 (SNX25) is a multifunctional intracellular trafficking protein of the sorting nexin family, characterized by a PX domain, a regulator of G-protein signaling (RGS) domain, and unique PXA/PXC domains that may function in lipid transfer. SNX25 critically regulates the turnover and lysosomal degradation of transforming growth factor beta (TGF-β) receptors, modulating cellular signaling and maintaining receptor balance. Structural studies indicate that SNX25 is anchored to the endoplasmic reticulum and facilitates lipid metabolism at various membrane contact sites. Dysregulation of SNX25-mediated receptor trafficking has been linked to epileptogenesis and may play a broader role in neurological diseases where TGF-β signaling is altered. SNX25 belongs to a subgroup of sorting nexins with an RGS domain and participates in cellular homeostasis and protein quality control. No established drugs currently target SNX25 directly, but its regulatory roles position it as a candidate for future therapeutic exploration.
