Overview

Sorting nexin-29 (SNX29) is a member of the sorting nexin (SNX) protein family, characterized by the presence of a phosphoinositide-binding PX domain that enables association with phosphatidylinositol-enriched membranes of the endosomal network[2]. SNX29 is categorized within the SNX-PX subfamily, which includes SNX proteins containing only the PX domain and is evolutionarily conserved among eukaryotes[2]. SNX proteins, in general, regulate endocytosis and the sorting of membrane protein cargoes through endosomal and lysosomal pathways, coordinating protein recycling or degradation. SNX29 itself has been associated with diverse biological processes and disease roles, such as growth and myogenesis in animals, psychiatric conditions (schizophrenia, autism), certain cancers, and immune function (IgA levels), mainly through observational and genetic studies[1]. In livestock, SNX29 gene copy number variation influences growth traits, making it an important molecular marker for breeding but not a direct therapeutic target. No drugs are known to directly interact with SNX29, nor is there an established therapeutic mechanism of action or notable human toxicity.
