Sorting nexin-32 is an endosomal trafficking protein, part of the SNX-BAR sub-family of sorting nexins. It contains a PX domain facilitating protein-protein interactions and a BAR domain involved in membrane curvature sensing and dimerization. SNX32 forms heterodimers with other SNX-BAR proteins such as SNX4, SNX1, and SNX6, contributing to the cargo selection and membrane recruitment to early endosomes. SNX32 is especially expressed in the brain, suggesting specialized roles in neurons. It interacts with a range of cargoes, notably transferrin receptor, CIMPR, and Basigin, impacting recycling and cell surface expression. SNX32 regulates neurite outgrowth and neuroglial coordination by trafficking essential proteins to the cell surface. Dysfunctional SNX32-mediated trafficking can impair neurite extension and cellular differentiation, linking it to processes relevant in neurobiology and possibly neurodegeneration[1][2][3].
